摘要:
目的 观察Ⅱ型胶原诱导及风寒湿、风湿热环境刺激对大鼠类风湿性关节炎(RA)模型的建立及血清免疫球蛋白G(IgG)含量、乳酸脱氢酶(LDH)活性的变化。方法 将40只SD大鼠随机分为空白组、Ⅱ型胶原组、Ⅱ型胶原+风寒湿组、Ⅱ型胶原+风湿热组,每组10只,雌雄各半。除空白组外,各组大鼠于实验当天在背部、尾根部皮内注射Ⅱ型胶原乳剂,第12天强化免疫。病证结合模型从造模第2天开始分别给予风寒湿或风湿热环境刺激,每次30 min,连续刺激28 d。其中,每3 d测定大鼠体质量变化,每2 d测定大鼠足趾肿胀度变化。至第28天处死,测定大鼠血清IgG含量和LDH活性。结果 造模后各模型组足趾肿胀度明显升高,至第16天达高峰,差异具有高度统计意义(P<0.01)。与空白组比较,各模型组大鼠血清IgG含量均有所升高,其中Ⅱ型胶原+风寒湿组升高较显著,差异有统计意义(P<0.05)。与空白组比较,LDH活性呈现2种趋势:Ⅱ型胶原组、Ⅱ型胶原+风寒湿组的LDH活性升高,差异有统计意义(P<0.05);Ⅱ型胶原+风湿热组的LDH活性降低,差异有统计意义(P<0.05),且Ⅱ型胶原+风寒湿组与Ⅱ型胶原+风湿热组的LDH活性差异有统计意义(P<0.05)。结论 Ⅱ型胶原加风寒湿、风湿热环境因素刺激,可以复制病证结合RA大鼠模型,这种模型在一定程度上可以模拟中医寒湿痹证和湿热痹证。
Abstract:
Objective To observe the change of the content of immunoglobulin G(IgG) and the activity of lactate dehydrogenase(LDH)in the serum of rat model with rheumatoid arthritis (RA). The rat models were induced by the cattle collagen Ⅱ and intervened by the environmental factors of wind cold dampness and rheumatic fever. Methods 40 SD rats, half-and-half male and female, were divided into four groups, 10 cases each group. They were normal group, collagen Ⅱgroup, collagen Ⅱcombined with wind cold dampness group, collagen Ⅱcombined with rheumatic fever group. Except the normal group, other 3 groups received intrademal injection with same dose of collagen Ⅱ. At the same time, the rat models combined with disease and syndrome were intervened by environmental factors of wind cold dampness and rheumatic fever from the 2nd day to the 28th day, lasting 30 minutes every day. Then the content of IgG and the activity of LDH were determined. Results The degrees of swelling in model groups were increased significantly and were at their peaks on the 16th day, there was a highly statistical significance(P<0.01). Compared with the normal group, the contents of IgG in each model group were increased, and the contents of IgG in collagen Ⅱcombined with wind cold dampness group were increased significantly(P<0.05), the activity of LDH in collagen Ⅱ, collagen Ⅱ combined with wind cold dampness group had an obvious rise, and collagen Ⅱcombined with rheumatic fever group had an obvious decrease, there was a statistical significance(P< 0.05). The activity of LDH in collagen Ⅱ combined with cold dampness group and collagen Ⅱcombined with rheumatic fever group had obvious difference(P<0.05). Conclusion The stimulation of collagen Ⅱcombined with environmental factor of wind cold dampness and rheumatic fever can copy RA rat model combining disease and syndrome. To a certain extent, the model may simulate cold dampness arthralgia syndrome and damp heat arthralgia syndrome of Chinese medicine.
