摘要:
目的:探讨通络益智散对拟血管性痴呆大鼠海马区兴奋性和抑制性氨基酸的影响。方法:选用改良的脑缺血再灌注配合腹腔注射硝普钠降压法复制血管性痴呆模型。将实验大鼠随机分为5组,即假手术组、模型组、尼莫地平组、通络益智散大剂量组、通络益智散小剂量组。给药3周后,取海马匀浆检测谷氨酸(Glu)含量,并观察海马区γ-氨基丁酸(GABA)的表达。结果:通络益智散可以显著降低拟血管性痴呆模型大鼠海马区Glu的含量、增强GABA的表达(P<0.01)。结论:通络益智散能够通过调节血管性痴呆大鼠海马区Glu和GABA的含量保护脑组织,这可能是其治疗血管性痴呆的作用机制之一。
Abstract:
Objective: To explore the effect of Tongluo Yizhi Powder on excitatory and inhibitory amino acids in hippocampus of rats with vascular dementia. Methods: Vascular dementia model was build by brain ischemia-reperfusion injury and coordinate the step-down method injecting sodium nitroprusside in abdominal cavity.The rats were randomly divided into sham group,model group,nimodiping group,Tongluo Yizhi Powder high and low dose group. After 3 weeks, the levels of Glu and expression of GABA in hippocampus were measured. Results: Tongluo Yizhi Powder could prominently increase the expression of GABA and decrease the level of Glu in hippocampus of rats with vascular dementia (P<0.01).Conclusion: Tongluo Yizhi Powder has the protective effects to the brain tissue of rats with vascular dementia by influence the level of Glu and GABA.It may be one of mechanism of Tongluo Yizhi Powder for vascular dementia.
