摘要:
目的 探讨玻璃苣醇提取物对慢性应激抑郁模型小鼠行为学的影响,为玻璃苣醇提取物抗抑郁作用的研究提供实验依据。方法 将48只KM小鼠随机分为玻璃苣醇提取物高、中、低剂量组,阳性对照组,正常对照组和模型组对照组6组,每组8只。玻璃苣醇提取物高、中、低剂量组分别给予玻璃苣醇提取物13.3 g/kg、6.65 g/kg、3.33 g/kg,阳性对照组给予盐酸氟西汀混悬液0.0026 g/kg,正常对照组和模型对照组每天给予同体积的蒸馏水,各组均灌胃给药,1次/d,连续给药21 d。同时,以慢性轻度不可预见性的7种应激因子刺激小鼠,建立慢性应激抑郁模型,并通过敞箱实验、自主活动测定、悬尾实验、糖水消耗实验、Y型迷宫实验观察玻璃苣醇提取物对慢性应激抑郁模型小鼠行为学的影响。结果 玻璃苣醇提物可使抑郁模型小鼠敞箱活动及自主活动次数增多、悬尾不动时间缩短、糖水摄入量增多以及学习记忆力提高(P<0.05或P<0.01)。结论 玻璃苣醇提取物可以改善慢性应激抑郁模型小鼠的行为活动,从而推测玻璃苣具有改善抑郁症状的作用。
Abstract:
Objective To investigate the antidepressant effect of ethanol extract of borage on chronic stress depression model mice. Methods 48 KM mice were randomly divided into 6 groups: 3 dose groups (high, me-dium, low), positive control group, normal control group and model group. Each group has 8 mice. The 3 dose groups were interfered with borage ethanol extract at the doses of 13.3 g/kg, 6.65 g/mg and 3.33 g/mg respectively. The positive group was given suspension of fluoxetine hydrochloride at 0.0026 g/kg. Same amount of distilled water was given to normal control group and model group. Intragastrical administration was used, one time a day for 21 days. At the same time, the mice were irritated by 7 kinds of chronic mild unpredictable stress (CUMS) and their behavioral activities were observed through various experiments. Results The ethanol extract of borage increased the mice’s box-opening and autonomic activities, shortened the time of tail suspension without moving. The mice’s sugar intake increased and their learning and memory were improved(P<0.05 or P<0.01). Conclusion The ethanol extract of borage can improve the behavioral activities of model mice with chronic stress depression and accordingly it is effective to relieve depression.
