摘要:
目的 研究石榴健胃片对脾肾阳虚型溃疡性结肠炎(SKD-UC)模型大鼠结肠肿瘤坏死因子-α(TNF-α)、白细胞介素(IL) -1β、IL-6和IL-10的影响。方法 采用15%大黄混悬液灌胃联合2,4,6-三硝基苯磺酸-乙醇溶液灌肠的病证结合法建立SKD-UC大鼠模型,将模型大鼠随机分为阳性组,模型组,石榴健胃片高、中、低剂量组,每组20只,另取20只作为空白组。造模成功后第2天开始进行药物干预,阳性组大鼠灌胃柳氮磺胺吡啶(SASP,0.36 g/kg),石榴健胃片高、中、低剂量组分别灌胃石榴健胃片混悬液2.4,1.2,0.6 g/kg,模型组和空白组灌胃等体积生理盐水,每天1次,连续21 d。末次给药后24 h处死大鼠,取结肠组织,采用逆转录-聚合酶链反应(RT-PCR)法检测结肠组织中IL-1β,IL-6,IL-10,TNF-α的基因表达。结果 与空白组比较,模型组大鼠结肠IL-1β,IL-6,TNF-α的mRNA相对表达量明显增高,IL-10的mRNA相对表达量明显降低,组间比较差异均有高度统计意义(P<0.01);与模型组比较,石榴健胃片高、中、低剂量组大鼠结肠IL-1β mRNA和TNF-α mRNA相对表达量明显降低(P<0.05或P<0.01),高、中剂量组IL-6 mRNA相对表达量明显降低(P<0.01),而高、中、低剂量组IL-10 mRNA相对表达量明显升高(P<0.05或P<0.01);与阳性组比较,石榴健胃片高剂量组大鼠结肠IL-1β,TNF-α和IL-6的mRNA相对表达量及中剂量组IL-1βmRNA相对表达量明显降低(P<0.05),高、中剂量组IL-10 mRNA相对表达量明显升高(P<0.05);各剂量组之间IL-1β,IL-6,TNF-α和IL-10的mRNA相对表达量比较差异均无统计意义(P>0.05)。结论 石榴健胃片对SKD-UC模型大鼠的结肠TNF-α,IL-1β,IL-6及IL-10的基因表达具有调节作用。
Abstract:
Objective To study the effects of Shiliu Jianwei Tablet on TNF-α, IL-1β, IL-6 and IL-10 in the model rats with ulcerative colitis (UC) due to yang-deficiency of spleen and kidney. Methods UC rat models were created by intragastric administration of rhubarb powder suspension (15%) combined with enema of 2, 4, 6-trinitro-benzene-sulfonic acid (TNBS)/ethanol. Wistar rats were randomly divided into blank group (normal saline), positive control group (SASP, 0.36 g/kg), model group (normal saline), high-(2.4 g/kg), middle-(1.2 g/kg) and low-dose (0.6 g/kg) groups interfered with Shiliu Jianwei Tablet, 20 in each group. The treatment was carried out once a day and lasted for 21 days in succession. Then the rats were put to death and colon tissues were extracted. The gene expressions of TNF-α, IL-1β, IL-6 and IL-10 were detected by RT-PCR. Results Compared with blank group, the mRNA of IL-1β, IL-6, TNF-α in model group increased obviously while IL-10 decreased (P<0.01). Compared with model group, the mRNA of IL-1β, TNF-α in the 3 dose-groups decreased (P< 0.05 or P<0.01); the mRNA of IL-6 in high- and middle-dose groups decreased obviously (P<0.01), while the mRNA of IL-10 in the 3 dose-groups increased (P<0.05 or P<0.01). Compared with positive control group, the mRNA of IL-1β, TNF-α and IL-6 in high-dose group as well as the mRNA of IL-1β in middle-dose group decreased (P<0.05), the mRNA of IL-10 in high- and middle-dose groups increased (P<0.05). There was no any statistic difference among dose-groups (P>0.05). Conclusion Shiliu Jianwei Tablet can play a regulation role on the gene expression of TNF-α, IL-1β, IL-6 and IL-10 in UC rats due to yang-deficiency of spleen and kidney.
