摘要:
目的 观察气候因素对Ⅱ型胶原蛋白所致类风湿关节炎(RA)模型大鼠关节炎症、免疫脏器指数及组织形态的影响。方法 将40只SD大鼠随机分为空白组、Ⅱ型胶原组、Ⅱ型胶原+人工风寒湿组(简称风寒湿组)、Ⅱ型胶原+人工风湿热组(简称风湿热组)4组, 每组10只,雌雄各半。空白组给予等量生理盐水,Ⅱ型胶原组皮下注射Ⅱ型胶原佐剂混合液,其余2组采用Ⅱ型胶原佐剂混合液诱导,外加人工气候因素制备RA大鼠模型。造模28 d后,比较各组大鼠的关节炎评分、胸腺指数、脾脏指数,并观察各组大鼠左后肢踝关节的组织形态改变。结果 与空白组比较,各模型组大鼠均出现了不同程度的足趾关节肿胀,关节炎评分均显著升高,差异有高度统计意义(P<0.01);各模型组脾脏指数均有所升高,其中风寒湿组与空白组比较差异有统计意义(P<0.05);组织形态观察,造模后,Ⅱ型胶原组、风寒湿组和风湿热组大鼠踝关节出现不同程度的滑膜增生、淋巴细胞浸润。结论 气候因素对Ⅱ型胶原蛋白所致RA模型大鼠关节炎症、组织形态具有一定的影响,能加重RA大鼠关节的病理损伤。
Abstract:
Objective To observe the influence of climatic factor on the plantar form and immune viscera index of model rats with rheumatoid arthritis (RA) induced by Ⅱ-collagen. Methods 40 SD rats(half male and half female)were randomly divided into four groups: normal control group, Ⅱ-collagen group, Ⅱ-collagen+simulated factor of wind-cold-dampness group (shortened as WCD group) and Ⅱ-collagen+simulated factor of rheumatic fever group (shortened as rheumatic fever group), ten in each group.The normal control group was given normal saline at same dose.Ⅱ-collagen group was intervened with the adjuvant mixture of Ⅱ-collagen. The other two groups were induced by adjuvant mixture of Ⅱ-collagen plus simulated climatic factors to get RA rat model. On 28th day after model making, arthritis score, thymus index and spleen index were compared, the histopathological changes of the left-behind foot plantar in all groups were observed. Results Compared with the normal control group, the other 3 model groups had plantar joint swelling in various degree, arthritis score increased significantly and there was a statistically significant difference (P<0.01). The spleen indexes of model groups increased, meanwhile the spleen index of WCD group was significantly greater than that of normal control group, there was a obvious difference (P<0.05). Ankle synovial hyperplasia and lymphocytic infiltration were found in three model groups. Conclusion Climatic factors have certain influence on the joint inflammation and morphology of RA rats induced byⅡ-collagen, also can aggravate the joint pathological lesion.
