摘要:
目的 研究归芪聪志汤对血管性痴呆(VD)模型大鼠大脑皮层线粒体功能的影响。方法 将120只大鼠称重,进行水迷宫测试,剔除不合格大鼠10只,使用区组随机法选出10只大鼠作为假手术组,其余大鼠采用两血管阻断改良法制备VD大鼠模型,经Morris水迷宫筛选出造模成功的大鼠,随机分为模型组,阳性药物对照组,归芪聪志汤高、中、低剂量组。各组给予相应的药物或蒸馏水灌胃4周后,HE染色观察海马神经细胞的损伤情况,采用比色法检测大鼠海马组织及大脑皮层其他部位线粒体复合物的活性及三磷酸腺苷(ATP)的含量。结果 与假手术组比较,模型组海马神经细胞损伤程度严重;与模型组比较,各治疗组海马神经细胞损伤程度均减轻,其中归芪聪志汤中剂量组海马神经细胞损伤程度改善最为明显。与假手术组比较,模型组海马组织及大脑皮层其他部位线粒体复合物Ⅰ~Ⅴ的活性及ATP含量均明显降低(P<0.05)。与模型组比较,阳性药物对照组与归芪聪志汤中剂量组大鼠海马组织线粒体复合物Ⅰ~Ⅴ的活性及ATP含量明显升高,除归芪聪志汤中剂量组的线粒体复合物Ⅰ与模型组比较差异无统计意义外,其他均有统计意义(P<0.05);阳性药物对照组与归芪聪志汤中剂量组大鼠大脑皮层其他部位线粒体复合物Ⅰ~Ⅴ的活性均升高,其中线粒体复合物Ⅰ,Ⅲ,Ⅳ,Ⅴ差异有统计意义(P<0.05),ATP含量均降低,但差异无统计意义(P>0.05)。与阳性药物对照组比较,归芪聪志汤中剂量组大鼠海马组织线粒体复合物Ⅴ的活性明显升高(P<0.05);归芪聪志汤中剂量组大脑皮层其他部位线粒体复合物Ⅰ活性明显升高,线粒体复合物Ⅴ活性明显降低,差异均有统计意义(P<0.05)。结论 归芪聪志汤能明显改善VD模型大鼠海马组织病变,其作用机制与改善海马组织和大脑皮层其他部位线粒体复合物Ⅰ~Ⅴ的活性及ATP含量有关。
Abstract:
Objective To study the effect of Guiqicongzhi Tang on mitochondrial function of cerebral cortex in rats with vascular dementia. Methods 120 rats were tested by water maze to get 110 qualified rats; 10 were divi-ded as sham operation group,others were established as VD models by improved two-vessel occlusion; qualified model rats were randomly divided into model group,positive control group,high-,middle-and low-dose of Guiqicong zhi Tang group. After 4-week drug administration and distilled water gavage,lesion of hippocampus neurons was observed by HE staining,activity of mitochondrial complex in rats’ hippocampal tissue and cerebral cortex as well as the content of ATP were determined by colorimetric method. Results Compared with sham operation group,the injury of hippocampus neurons in model group was worse; compared with model group,the injury in all treatment groups were eased,especially obvious in middle-dose group. Compared with sham operation group,the activity of mitochondrial complex Ⅰ to Ⅴ and the content of ATP in model group significantly decreased (P<0.05); compared with model group,the activity of mitochondrial complex Ⅰ to Ⅴ and the content of ATP in middle-dose group and positive control group significantly increased,except complex Ⅰ,the differences were significant (P<0.05). The activity of mitochondrial complex Ⅰ to Ⅴ in middle-dose group and positive control group significantly increased,the differences of Ⅰ,Ⅲ,Ⅳ,Ⅴ were significant,while the content of ATP decreased without any statistical significance (P>0.05). Compared with positive control group,activity of mitochondrial complex Ⅴ in hippocampal tissue of middle-dose group increased (P<0.05). Activity of mitochondrial complex Ⅰ in cerebral cortex increased,while mitochondrial complex Ⅴ decreased,the differences were significant (P<0.05). Conclusion Guiqicongzhi Tang can obviously improve the lesions of hippocampus in VD model rats and the mechanisms may be associated with the improvement on activity of mitochondrial complex Ⅰ to Ⅴ and the content of ATP in hippocampus tissues and other parts of cortex.
