摘要:
目的 观察加味蒌石汤对肾阴虚型多囊卵巢综合征(PCOS)模型小鼠卵巢功能的保护作用,并初步探究其作用机制。方法 将54只小鼠随机分为正常对照组,模型组,达英-35组,加味蒌石汤高、中、低剂量组6组,每组9只。除正常对照组外,其余各组均采用脱氢表雄酮(DHEA)联合甲状腺素及利血平法制备肾阴虚型PCOS小鼠模型,各组给予相应药物进行预防性治疗。观察小鼠阴道脱落细胞和卵巢组织形态学改变,称量小鼠体质量及卵巢质量,计算卵巢指数,采用酶联免疫吸附法测定小鼠血清性激素[血清睾酮(T)、雌二醇(E2)、促卵泡刺激素(FSH)和黄体生成素(LH)]水平,抗苗勒氏管激素(AMH)、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)水平及cAMP/cGMP比值,从而评估加味蒌石汤对肾阴虚型PCOS模型小鼠的影响。结果 与正常对照组比较,模型组小鼠体质量增量显著降低(P<0.05),动情周期紊乱,卵巢湿重及卵巢指数均显著升高(P<0.05),血清T,LH,cAMP,AMH水平及cAMP/cGMP比值均显著升高(P<0.01),血清E2,FSH,cGMP水平均显著降低(P<0.01);与模型组比较,加味蒌石汤中、高剂量组及达英-35组小鼠体质量增量均显著升高(P<0.05),卵巢湿重及卵巢指数均显著降低(P<0.05),各药物组动情周期逐渐恢复正常,血清T,LH,cAMP,AMH水平及cAMP/cGMP比值均显著降低,血清E2,FSH,cGMP水平均显著升高(P<0.05或P<0.01);各药物组之间比较,加味蒌石汤高、中剂量组及达英-35组对以上指标的改善作用显著优于加味蒌石汤低剂量组,差异均有统计意义(P<0.05)。结论 加味蒌石汤能有效改善血清性激素T,E2,LH,FSH水平,且随药物剂量的增高效果越好;加味蒌石汤可通过调节AMH,cAMP水平和cAMP/cGMP比值改善PCOS模型小鼠肾阴虚症状,促进卵泡发育。
Abstract:
Objective To investigate the protective effect of Jiawei Loushi Tang on ovarian function of model mice with polycystic ovary syndrome (PCOS) due to deficiency of kidney-yin and explore its mechanism of action. Methods 54 mice were randomly divided into 6 groups:normal control group,model group,Diane-35 group,high-,medium-and low-dose Jiawei Loushi Tang group,9 mice each. Except normal control group,other groups were all modeled into PCOS rats with deficiency of kidney-yin syndrome. Exfoliated cells of vagina and ovarian pathological morphology were observed,body and ovary were weighed;ovary index was calculated;serum sex hormones,anti-Mullerian hormone (AMH),cyclic adenosine monophosphate (cAMP),cyclic guanosine monophosphate (cGMP) and the ratio between cAMP and cGMP were determined by ELISA to evaluate the protective effect. Results Compared with normal control group,the mice’s body weight in model group decreased (P<0.05),estrous cycle was in disorder,ovarian wet weight and ovarian index increased obviously (P<0.05),the serum levels of T,LH,cAMP,AMH and the ratio of cAMP and cGMP increased (P<0.01),while levels of E2,FSH,cGMP decreased (P<0.01). Compared with model group,the weight gain in medium-,high-dose group and Diane-35 group increased (P<0.05),ovarian wet weight and ovarian index decreased (P<0.05),the estrous cycle gradually returned to normal,the serum levels of T,LH,cAMP,AMH and the ratio of cAMP and cGMP decreased while the le-vels of E2,FSH,cGMP increased (P<0.05 or P<0.01). Compared among all drug groups,improvement of above indexes in high-,medium-dose and Diane-35 group was superior to low-dose group,the difference was statically significant (P<0.05). Conclusion Jiawei Loushi Tang can effectively improve the serum levels of sex hormones T,E2,LH,FSH and the effect goes better with the rise of dose. It also can improve the deficiency of kidney-yin symptoms and promote follicular growth by adjusting the levels of AMH,cAMP and the ratio of cAMP and cAMP.
