摘要:
目的 观察槲皮素联合伊马替尼对人胃癌SNU-1细胞生长和细胞凋亡的影响。方法 以12.5~200.0 μmol/L槲皮素和0.125~2.000 μmol/L伊马替尼单药以及二者联合用药处理细胞24 h,用噻唑蓝(MTT)法检测细胞增殖,Hoechst 33342染色观察细胞核变化,流式细胞仪检测细胞凋亡情况;用蛋白印迹法检测相关蛋白的表达情况。结果 槲皮素和伊马替尼单药均能使细胞活力降低,0.250 μmol/L伊马替尼和25.0 μmol/L槲皮素联合用药对细胞活力具有显著抑制作用,差异有高度统计意义(P<0.01),且可诱导人胃癌SNU-1细胞凋亡;二者联合处理还可以显著诱导半胱氨酸蛋白酶-9(Caspase-9)和半胱氨酸蛋白酶-3(Caspase-3)的上调,差异有高度统计意义(P<0.01)。结论 槲皮素联合伊马替尼能抑制胃癌SNU-1细胞的生长并诱导细胞凋亡,其作用机制主要是通过上调Caspase-9和Caspase-3的表达从而启动细胞凋亡来实现的。
Abstract:
Objective To investigate the effect of combined use of quercetin and imatinib on proliferation and apoptosis of SNU-1 cells. Methods SNU-1 cells were treated by different concentration of quercetin (12.5 to 200.0 μmol/L) and imatinib (0.125 to 2.000 μmol/L) alone or both for 24 hours. Cell proliferation was assayed by MTT method. Hoechst 33342 staining method was used to observe the change of cell nucleus. Apoptosis was detected by flow cytometry and expression of caspase-8,9 and 3 protein were measured by westernblotting. Results Single or combined use of quercetin and imatinib induced cell apoptosis and up-regulated the expression of caspase 9 and 3 protein in SNU-1 cells,especially did in combination. The differences were statistically significant (P<0.01). Conclusion Quercetin combining imatinib can synergistically inhibit cell proliferation and induce cell apoptosis probably by up-regulating the expression of caspase-9 and 3 protein.
